Travelling from Kalyani Ngr to Wakad in a car: Which is the best route to take and how much approx. time will it take?

The standard route via Sancheti flyover - University Road is quite busy and might take your longer during busy commuting hours.

One alternative road is to go via Yerawada - Alandi Road - Bombay Sappers - Holkar Bridge - Old MumbaiPune Highway - Dapodi. Check this out on google maps.

this altenative route does work out better bcoz the traffic is not as much as the sancheti-unv rd.

I think both routes will take you about 35 minutes (if you don't hit a traffic jam), and maybe 10 to 15 minutes more if you do hit a traffic jam.
